Membrane transporters can be major determinants of the pharmacokinetic, safety and efficacy profiles of drugs. Monolayer assays using transfected MDCKII-MDR1/P-gp or BCRP (ABCG2) cells have been widely recognized tools to study the interaction of drugs with transporters. SOLVO Biotecnology and Readycell introduce PreadyPort™, the Ready-to-use Transfected MDCKII Monolayer Kits containing differentiated MDCKII cells expressing the selected drug transporter on 24- or 96-well cell culture insert plates.

PreadyPort™-MDR1 Kit contains 24 & 96 insert-integrated plates with differentiated MDCKII cells expressing MDR1, as well as the parental cell line. The first member of the PreadyPort series, the PreadyPort™-MDR1 Kit is revolutionary in providing a ready-to-use tool for MDCKII-MDR1 monolayer assays. The kit will allow researchers to perform MDR1 interaction studies on monolayers without worrying about cell-line licensing and culturing.

The patented shipping medium allows storage and transportation at room temperature, without altering cell functional properties.

Stability of PreadyPort™-MDR1 barrier properties under shipping conditions

The barrier stability of the PreadyPort-MDR1 kit during shipping and storage is presented above. The 11-day MDCKII-WT and MDR1 monolayers were maintained in shipping medium for 1, 2, 3 and 4 days, then their barrier status was evaluated by TEER (trans-epithelial electrical resistance) measurement before applying and 24 hours after removing the shipping medium. Cell monolayers showed no changes in barrier properties up to 4 days in the presence of shipping medium indicating that PreadyPort-MDR1 can be stored and transported at room temperature up to 4 days without loss of its barriers functions.

  • Functional Stability of PreadyPort-MDR1 in 24 & 96-insert well plate formats

    Pgp/MDR1-mediated digoxin transport was determined from three independent experiments at day 12 of culture in both, 24 and 96-insert integrated plates formats. Transport of digoxin was specifically inhibited by PSC833 Pgp inhibitor.

High Flexibility of PreadyPort-BCRP

    The shipping medium was maintained for 4 days at room temperature. After removed, TEER was measured after 3 (D11) and 7 (D15) days of standard culture conditions. Results represent the average of three independent experiments.

  • Functional Stability of PreadyPort-BCRP in 24-insert well plate format

    BCRP-mediated prazosin transport was determined from three independent experiments at day 12 of culture in 24-insert integrated plate format. Transport of prazosin was specifically inhibited by Ko134 BCRP inhibitor. No transport was seen in MDCKII-wt cells.

  • Papp values obtained with PreadyPortTM-BCRP after the incubation of different BCRP substrates and the specific BCRP inhibitor Ko143
